ELISA: To detect antibodies to viruses, viral protein is linked to the plastic support, and then the clinical specimen is added. If antibodies against the virus are present in the specimen, they will bind to the immobilized antigen. The bound antibodies are then detected by using a second antibody that binds to the first antibody.
MAT: Serial dilution of serum kept in contact with an equal volume of a well grown suspension of leptospires at a certain temperature for a certain period of time and read microscopically by estimating 50% agglutination as the end point titer of he reaction mixture.
