Summary of Results:

The overall objective of my laboratory is to understand better the mechanisms of follicle cell differentiation. During follicle development, there is significant growth of the follicle and proliferation and differentiation of granulosa cells.  We are currently studying roles of members of the fibroblast growth factor (FGF) family. This large family contains 22 peptides and 7 receptors (which result from alternative splicing of 4 FGFR genes).  We have described the expression of a number of these FGFR splice variants in the bovine follicle, and we are determining which FGFs play important roles in follicle development. We have shown that FGF10 mRNA and protein are present in the follicle. FGF10 is expressed by the theca cells and the oocyte, but not by granulosa cells. Thus there is a potential for paracrine signaling to granulosa cells from theca cells or the oocyte.  We tested this by culturing granulosa cells with FGF10, and found that FGF10 significantly inhibits granulosa cell steroidogenesis. 

We are extending this work to look at another FGF, FGF18. This factor forms part of a subfamily that includes FGF8, and FGF8 is a oocyte-specific factor in mice, and we have detected FGF8 mRNA in cattle. FGF18 has been described in oocytes in mice, although we failed to detect FGF18 in bovine oocytes. However, it is detected in theca cells and in granulosa cell of atretic follicles. We are now investigating the actions of FGF18 in granulosa cell culture.
